The process of antibody chimerisation results in an antibody where the variable regions of your monoclonal antibody have been grafted onto a suitable framework. Typically this is done when your antibody is of murine origin and you wish for it to resemble a human antibody. Antibody chimerisation is the first step towards engineering a fully humanised antibody by CDR grafting.
The chimerisation, as with isotype switching, is a rather straight forward procedure. It requires cloning and sequencing of variable region cDNA, grafting the sequence information onto the required framework, synthesising the resulting gene and finally producing the recombinant antibody in a mammalian expression system.
