We offer custom production of methylated peptides. Methylation is a post translational modification, where typically one or more hydrogens in Arginine or Lysine are replaced by a methyl group. Methylated Arginine is either mono-methylated or di-methylated - symmetric or asymmetric - while Lysines are mono-, di- or tri-methylated.
The incorporation of methylated residues is typically done by manual synthesis due to the comparatively high reagent cost, but other than that the same strategy applies as for the synthesis of any other peptide. The main issue here is the quality of the reagents. Using high quality reagents is important for any peptide synthesis, but in this case it becomes truly pivotal.
For example, Lysine can be either mono-, di- or tri-methylated. Let's assume that you would like your peptide to have a tri-methyl Lysine. If the Lys(Me3) building block is contaminated by Lys(Me) and Lys(Me2), it will generally not be possible to separate the three peptide species in the HPLC purification procedure. Thus, the quality of the Lys(Me3) building block is essential for the successful production of your peptide.
While the preparative HPLC cannot discriminate - as an example - between peptide species containing di- and tri-methylated Lysine, the difference in mass is however recorded in the mass spectrum analysis. Although MS records relative intensity and cannot be used as a reliable measure of the level of impurities, a sensitive high end quadrupole MS does offer what could be considered semi-quantitative results. The QMS also allows for the in-house QC of synthesis reagents that are used for making methylated peptides.
