There are individual differences in the animals, not only in how they will respond to your immunogen, but also in what is already present their system. Screening pre-immune samples from a number of individuals is a good idea in order to select one which will give the lowest background in your application.
Typically you will have to test 3-5 individuals per intended immunisation. In most cases this number suffices, but there have been cases where 10 animals needed to be tested before finding a suitable candidate.
However, if you are planning on using material that has been antigen specific affinity purified – especially if the antigen is a synthetic peptide - then it is questionable if the screening is meaningful.
